Pansol 20 Tablet

Composition

Pansol 20 Tablet: Each delayed-release tablet contains Pantoprazole Sodium sesquihydrate USP equivalent to Pantoprazole 20 mg.

Pharmacology

Pantoprazole is a proton pump inhibitor that suppresses gastric acid secretion by blocking the hydrogen-potassium ATPase enzyme system in gastric parietal cells.

It is well absorbed with a bioavailability of ~77% and peak concentration (~2.5 µg/ml) occurring approximately 2.5 hours after administration. The drug is extensively metabolized in the liver and mainly excreted via urine (71%) and feces (18%).

Food may delay absorption, but does not alter the extent, so it can be taken with or without food.

Indications

  • Benign gastric ulcer
  • Duodenal ulcer
  • Gastroesophageal Reflux Disease (GERD)
  • NSAID-induced peptic ulcer
  • Acid hypersecretory conditions (e.g. Zollinger-Ellison syndrome)
  • Helicobacter pylori eradication (with antibiotics)
  • Ulcers resistant to H2-receptor antagonists

Dosage and Administration

  • Benign gastric ulcer: 40 mg once daily for 4–8 weeks
  • Duodenal ulcer: 40 mg once daily for 2–4 weeks
  • GERD: 20–40 mg once daily for 4–8 weeks
  • NSAID-induced ulcers: 20 mg once daily
  • Zollinger-Ellison syndrome: Initially 80 mg/day, adjusted per response; doses above 80 mg should be divided
  • H. pylori eradication: 40 mg twice daily with antibiotics
  • H2-antagonist resistant ulcers: 40 mg once daily for 8 weeks
  • Maintenance: 20 mg once daily; may increase to 40 mg if symptoms recur

Contraindications

Contraindicated in patients with known hypersensitivity to pantoprazole or any component of the formulation.

Precautions

  • Do not split, crush, or chew the tablet; swallow whole.
  • Concomitant use of antacids does not affect absorption.

Side Effects

Common: Headache, diarrhea

Rare: Abdominal pain, flatulence, rash, insomnia, hyperglycemia

Drug Interactions

No dose adjustments are needed when used with theophylline, diazepam, digoxin, antibiotics (clarithromycin, metronidazole, amoxicillin), oral contraceptives, etc.

May affect absorption of drugs dependent on low gastric pH (e.g. ketoconazole, iron salts, ampicillin esters).

Use in Pregnancy & Lactation

Animal studies show no harm to the fetus. Human data is limited—use only if clearly needed.

Pantoprazole is excreted in breast milk. Consider discontinuing breastfeeding or the drug based on benefit-risk evaluation.

Storage

Store below 30°C. Protect from light and moisture. Keep out of reach of children.
